The method of inoculation and effectiveness of oat (Avena sativa L.) and maize (Zea mays L.) rhizosphere settling by bacteria of the Azospirillum genus
Summary.

The possibility of utilisation of Azospirillum bacteria as a factor exerting a beneficial impact on yields of crop plants requires, among others, elaboration of inoculation methods with these bacteria ensuring effective colonisation of the plant root zone area and, at the same time, possible for the application in agricultural practice. The aim of the research project was to recognise the settling effectiveness of oats and maize rhizosphere and rhizoplane with bacteria of the Azospirillum genus depending on the method of inoculation with the A. brasilense strain. The following four combinations were employed in the performed pot experiment: pre-sowing soil inoculation, kernel inoculation, post-emergence soil inoculation and simultaneous application of all these three inoculation variants. The 65B strain isolated at the Section of Microbiology of Institute of Soil Science and Plant Cultivation – National Research Institute in Puławy was applied. Various analyses aiming to determine bacteria of the Azospirillum genus in individual experimental combinations were carried out throughout the vegetation period. Simultaneously, chlorophyll concentrations in plant leaf blades were controlled and soil nitrogenase activity was determined by the acetylene method. The results of the performed investigations show that the least effective method of colonisation initiation of oats and maize rhizosphere by A. brasilense was kernel inoculation. On the other hand, introduction of these bacteria into the soil, both pre-sowing and post-emergence, increased counts of bacteria of the Azospirillum genus in the rhizosphere of oats and maize. Increased numbers of A. brasilense following inoculation failed to find its reflection in plant vitality expressed by chlorophyll concentration in their leaf blades. Although in the case of oats the relationship between the very fact of inoculation and chlorophyll concentration was fairly clear, in the case of maize, it was barely noticeable.
